In the past six years, the number of craft breweries (The Brewers Association, based in Boulder, Colo., defines craft breweries as those that produce less than 6 million barrels per year.) and brewpubs has tripled to reach 5,525 breweries, according to the
ATTB report.
We wanted to clone the construct mRFP-EGFP-rLC3 into the pUAST attB vector.
The cells were transfected with the pUAST attB vector and the actin-Gal4 driver, to drive the expression of the mRFP-EGFP-rLc3 construct.
The pUAST attB plasmid was kindly supplied by Konrad Basler, while the mammalian plasmid containing the mRFP-EGFP-rLC3 marker insert was kindly supplied by Prof.
Using Gateway[R] Recombination Cloning Technology [21], the specific fragments of EF1[alpha]-hTERT containing the
attB adaptor were PCR amplified using Phusion[R] high-fidelity DNA polymerase (New England BioLabs, Singapore).
This instrumentation is: movement I: SATB; II: SAAB; III: SAAA; IV: ATTB; and V: SATB.
While It's Lovely was originally scored for AATB quartet and Yuppieville Rodeo calls for ATTB quartet, both are actually published with extra parts to allow for performance by the standard SATB instrumentation, which is employed for Kentucky Roastup.
Perhaps the least successful of these transposed works is Nicholas Gombert's O domina mundi (NDCE 59), which Buxton moves up a perfect fifth instead of leaving it as a low-voice piece (
ATTB) and transposing it up the more practical distance of a major third.
All settings employ similar compositional techniques, utilize the identical eight-voice ensembles of CATB and
ATTB notated in the same set of standard clefs, and treat the relationship between the two choirs in a similar manner.
