Thiazides and similar produce and increase in sodium and lithium reabsorption in the proximal convoluted tubule (where their reabsorption is more relevant).
Supportive findings include the expression of proximal convoluted tubule markers (CD10 and alpha-methylacyl-CoA racemase; AMACR), distal nephron proteins (parvalbumin, high-molecular-weight CK, and CD19) [1, 6], and the detection of intercalated cells and cells similar to those in the proximal tubule using electron microscopy [1, 7, 8].
Using this mouse model, we have demonstrated marked differences in renal structure and function between the Hp 1-1 and Hp 2-2 DM mice; Hp 2-2 DM mice had a significant increase of iron deposits in the lysosomes of the kidney proximal convoluted tubule cells (PCT) with lysosomal and renal damage as well as increased features of glomerular disease characteristic of early human DN [7, 13, 14].
The immunoreactivity was weak in the glomerulus and the peritubular capillary (Pcap), but no staining was noted in the proximal convoluted tubule (PCT), the distal convoluted tubule (DCT), or the CCD.
